Preparation of expression vectors for the production of ZIKA and Dengue NS3 helicase

Zika and Dengue viruses have spread, due to globalisation, to all continents which lie at least in part in the subtropic and tropic climatic zones. This spread of these viruses is a reason of an increasing number of severe diseases caused by them. New drugs, which would be effective against these infections, could be an answer to this challenge. Various viral proteins, among them also viral helicase, which is the topic of this bachelor thesis, can be a suitable drug target. The task was to prepare expression constructs for production of recombinant helicases of Zika and Dengue viruses via the suitable bacterial strain Escherichia coli. Several constructs derived from plasmid pET-16b were prepared with inserted helicase of Zika and Dengue viruses. One of them was used for the preparation of recombinant purified helicase of Zika virus, that will be used for further research. Computational modeling of complexes consisting of the Zika virus RNA-dependent RNA polymerase and NTP analogs

The recently published crystallographic structure of the Zika virus RNA-dependent RNA polymerase (RdRp) allows to produce molecular dyna- mics (MD) simulations of this enzyme that can help in the rational design of its inhibitors - potential chemotherapeutics. In the theoretical part of the thesis are first described basic principles of MD simulations and the method of calculation of binding free energy. This is followed by a brief introduction into the structure of nucleic acids, proteins, RNA polymerases and into mechanisms of expression of genetic information. In the practical part are presented the alchemical trans- formations of ADP to its analogues, which were recently synthesized at ÚOCHB AV ČR. Transformations are performed first in the water envelope and then in the active site of HCV RdRp. Additionally, the conformation of the Zika virus RdRp is modified to match the hepatitis C virus polymerase conformation that can accommodate nucleic acids. Finally, MD simulations of the ADP analogues in the active site of the Zika virus polymerase are produced.
The Role of the Media in Construction of Health Risks in Czech Republic on the Example of Ziza Virus

This Master's thesis identifies the characteristics of the Czech media in the context of the media construction of health risks - the Zika virus, focusing on the printed media. Research by T. Knířová (2011), comparing the construction of swine flu risk between the British daily newspaper The Times and the Czech daily MF Dnes, and the analysis of the avian influenza, mad cow disease and West Nile fever in The New York Times (Shih, Wijaya, Brossard, 2008) are the initial works. The thesis uses a method of quantitative content analysis extended by an interpretative text reading. The risk of viral infection is analyzed with regard to concepts of agenda-setting, framing, and issue-attention cycles.
The social impact of the media coverage of world epidemics (exemplified on ebola and zika viruses)

This bachelor thesis focuses on the media influence on society during Ebola epidemic (in 2014-2016) and Zika epidemic (in 2016). The theoretical part describes the basic concepts and outlines the progress of both epidemics. Then presents a theoretical basis for the issue of the influence of media messages on the audience, society and the pharmaceutical industry. This part also briefly discusses the adaptation of reporting about diseases in the selected ethical codes (The Czech Television Code, BBC Editorial Guidelines and The Code of Association of Television Organizations CR). In the practical part, the findings from the theoretical part are applied in the case study, where the audio-visual news from The Czech television and Television Nova during Ebola and Zika epidemics are analysed. The thesis notices frequency of the news during these periods, the character of the news, language usage and the guests who appear in the broadcasting. The analysis is accompanied by a description of the editorial work in both televisions. Subsequently, based on the results of opinion polls, there are chosen the most visible effects, that the media coverage caused on the audience.
